What Is MGF Peptide?

MGF (mechano growth factor) is an alternatively spliced form of insulin-like growth factor 1 (IGF-1). In humans, it is also known as IGF-1Ec. MGF belongs to the IGF-1 family of peptide growth factors and contains a distinct E-domain produced through alternative splicing of the IGF-1 gene.

The term MGF is used in scientific literature to describe this IGF-1 splice variant and, in some studies, the corresponding MGF-derived peptide. Its classification is therefore based on its relationship to the IGF-1 gene and protein family rather than being a separate gene product.

MGF-Ct24E is a 24-amino-acid peptide corresponding to the C-terminal region of the human IGF-1Ec/MGF E-domain. Human IGF-1Ec is produced through alternative splicing of the IGF-1 gene and contains a unique C-terminal sequence resulting from this splicing pattern.

The synthetic MGF peptide is therefore a defined short peptide sequence derived from the C-terminal region of IGF-1Ec, rather than the complete IGF-1 protein. Published studies have specifically described and investigated this 24-amino-acid MGF-Ct24E sequence.

Mechanism of Action Under Investigation

Published studies indicate that the cellular activity of MGF-Ct24E does not appear to involve the IGF-1 receptor (IGF-1R). Experiments with human myogenic precursor cells found that responses to the synthetic peptide were not dependent on IGF-1R. A specific receptor through which MGF-Ct24E acts has not been identified.

Downstream signaling investigated in experimental studies includes the MAPK/ERK pathway. MGF-Ct24E increased ERK1/2 phosphorylation in osteoblasts, and inhibition of the MAPK/ERK1/2 pathway reduced the cellular response. These findings support involvement of ERK-related signaling in the experimental response to the peptide.

The upstream molecular interaction remains unresolved. Published research has not established a specific receptor for MGF-Ct24E or a complete signaling pathway connecting the peptide to ERK activation. Therefore, the molecular mechanism remains under investigation

Scientific Research Context

MGF has been studied in the scientific literature for several decades. The evidence base includes in vitro cell studies, preclinical animal research, and a smaller number of human studies. Research has largely centered on skeletal muscle and related tissues. Most of the available evidence remains preclinical, while human research represents a more limited part of the literature. Overall, the research has progressed from cellular and animal studies toward a smaller body of human investigation.

06

MGF Biology and Biological Pathways

01

Primary Biological Function

MGF is an alternatively spliced IGF-1 isoform associated with mechanical stimulation and tissue-specific expression. Research has examined its biological role mainly in skeletal muscle, where MGF expression changes in response to exercise and mechanical loading. Researchers have also investigated MGF expression in bone, cartilage, tendon, and other tissues.

02

Relevant Metabolic or Signaling Pathways

MGF-related research has examined signaling associated with cell proliferation, differentiation, and cellular responses to mechanical stimuli. Researchers have investigated the ERK1/2 pathway in osteoblast studies involving MGF-Ct24E. Research has also examined changes in IGF-1 splice-variant expression as part of the broader IGF-1 signaling system.

03

Tissue-Specific Biology

MGF expression has been investigated mainly in skeletal muscle, where studies have examined its expression following exercise and muscle injury. Research has also identified MGF expression in bone, growth-plate cartilage, and tendon, as well as in cardiac and brain tissues under specific experimental conditions. These findings show that MGF expression is not limited to skeletal muscle and can vary between tissues.

04

Experimental Biomarkers

Researchers measure MGF mRNA and protein expression to assess MGF expression. Human skeletal muscle studies measure MGF mRNA using RT-PCR, while other studies measure MGF protein levels. Researchers also measure ERK1/2 phosphorylation in cellular studies of MGF-Ct24E.

05

Laboratory Investigation Methods

MGF has been investigated using muscle biopsies, cell culture and PCR-based gene-expression analysis. Human muscle studies have used biopsy samples with real-time quantitative RT-PCR to measure MGF mRNA expression. Cell-based studies have used cultured muscle and other cell types to examine MGF-related responses. Researchers have also assessed protein expression using immunohistochemistry and Western blot analysis.

07

Experimental Models Used in Published Research

01

Cell Culture Models

Published MGF studies have used C2C12 myoblasts, primary mouse muscle stem cells, primary human myoblasts, and osteoblast cultures. These models are limited because isolated cells do not reproduce the cellular and physiological complexity of intact tissue.

02

Three-Dimensional Muscle Models

Published MGF research has used three-dimensional C2C12 skeletal muscle cultures embedded in collagen constructs. The model remains an in vitro system. So it does not reproduce the full cellular and physiological complexity of intact skeletal muscle.

03

Mouse Models

MGF-Ct24E has been studied in mouse models involving transplanted human myogenic precursor cells. These models are limited by species differences and by the differences between an experimental transplantation system and normal human physiology.

04

Rabbit Models

Published MGF-Ct24E research has used a rabbit segmental bone-defect model involving a surgically created radial bone defect. Its main limitations are the species difference between rabbits and humans and the fact that the experimentally created defect does not reproduce the full complexity of human bone conditions.

05

Human Tissue Models

Published MGF research has used human skeletal muscle biopsy samples and primary human myogenic cells. Their main limitations are limited tissue sampling and the inability of isolated cells or biopsy samples to reproduce the full physiology of intact human muscle.

08

09

Limitations of Current Research

01

Evidence Is Predominantly Preclinical

MGF research remains predominantly preclinical. Much of the literature comes from cell based and animal studies, with relatively limited human research.

02

Human Clinical Evidence

Controlled human clinical evidence remains limited. Human studies have mainly examined MGF/IGF-1Ec expression in skeletal muscle rather than evaluating MGF-Ct24E in controlled clinical studies.

03

Species Differences

MGF-related IGF-1 splice variants differ between species. Human IGF-1Ec and rodent IGF-1Eb have different sequences, which limits direct comparison of MGF findings across species.

04

Study Design Limitations

Studies have used different MGF molecular forms and experimental conditions. This makes it difficult to compare findings across experiments, particularly when studies treat the endogenous splice variant and synthetic MGF-derived peptides as equivalent.

05

Independent Replication

Independent replication remains limited because published studies have reported different findings for MGF peptide activity. A later study did not reproduce earlier reported effects on myoblast proliferation or differentiation in human and mouse muscle-cell systems. This inconsistency makes the reported biological effects of MGF peptide less certain.

06

Long-Term Evidence

Long-term evidence remains limited. Most MGF studies have examined short-term changes in MGF expression or cellular responses, while relatively few have followed MGF related effects over extended periods. As a result, the longer-term biological effects of MGF remain insufficiently characterized.

Analytical Documentation

HPLC can assess peptide purity by separating the target peptide from related impurities and other sample components. It provides chromatographic information that helps characterize the composition of a sample.

LC-MS can provide molecular-mass information and support peptide identity by comparing the measured mass with the expected mass. NMR can provide additional structural information when the sample and analytical purpose make this technique appropriate.

A Certificate of Analysis (CoA) records batch-specific analytical results, such as identity and purity, together with the methods used for testing. This documentation records the analytical characteristics of the material used in laboratory research.

Analytical verification helps establish the identity and quality of research material before experimental use. Analytical documentation verifies product identity and quality, not biological performance.

11

Storage and Laboratory Handling

Store the material at 2–8°C in its original container. Keep the container closed when not in use, and protect the material from moisture, heat, and direct light.

Qualified laboratory personnel should handle the material in a clean laboratory environment. Use clean equipment and work surfaces, and follow standard laboratory practices to prevent contamination during storage and handling.

12

Frequently asked questions

MGF, or mechano growth factor, is the name commonly used for the IGF-1Ec splice variant in humans. Alternative splicing of the IGF-1 gene gives IGF-1Ec a distinct C-terminal E-domain. The term MGF is also used for the synthetic 24-amino-acid peptide corresponding to the C-terminal region of this E-domain. Because the literature uses MGF for both the splice variant and the synthetic peptide, the specific molecular form should be identified when discussing research findings.
